I made a small paint booth 18"x18" to spray on lacquer finish.
My question is can i connect my dust collector to the back to remove the overspray?

Not a good idea to clog the ducting system. You also do not want explosive fumes in a collection system like that. ( sparks is a bad thing) The pros use explosion proof fans and filtering system.

i see no reason why you can't. however i would not as i feel the lacquer would harden and seal off the pores of the filter/bag. a better solution might be to use a bathroom exhaust type fan and vent it outside being careful of the location. ie: where vehicles are parked (yours or your neighbors). just my $.02

Spray lacquer can be very explosive , especially as a fine mist as in the over spray . Be very careful about using any kind of exhaust fan because DC motors with brushes create sparks and some AC motors can get hot enough ignite the vapors . Use ONLY an exhaust approved for use in explosive situations .
